HMG-CoA reductase (pSer871) antibody
Background:HMG-CoA reductase is the rate-limiting enzyme for cholesterol synthesis and is regulated via a negative feedback mechanism mediated by sterols and non-sterol metabolites derived from mevalonate, the product of the reaction catalyzed by reductase. Normally in mammalian cells this enzyme is suppressed by cholesterol derived from the internalization and degradation of low density lipoprotein (LDL) via the LDL receptor. Competitive inhibitors of the reductase induce the expression of LDL receptors in the liver, which in turn increases the catabolism of plasma LDL and lowers the plasma concentration of cholesterol, an important determinant of atherosclerosis.
Clarke and Hardie identified Ser871 of HMG CoA reductase as a substrate of AMP-activated kinase. The residue is highly conserved in rodents and man.
Description: Rabbit polyclonal to HMG-CoA reductase (pSer871)
Immunogen: A synthesized phosphopeptide derived from HMG-CoA reductase around the phosphorylation site of serine (N-R-SP-K-I).
Specificity : ·Reacts with Mouse, Rat and Hamster.Recognizes HMG-CoA Reductase when phosphorylated on Ser872(Human).
· Use for Western blot, Immunohistochemistryand ELISA.
Purity: Immunogen affinity purified. Double affinity purification over both a nonphospho- peptide column, followed by phopshorylated peptide column.
Application: ·Western blot:1/500-1000. Predicted molecular weight: 98 kDa.
·Immunohistochemistry:1/200-500.
·Optimal working dilutions must be determined by the end user.
